Effects of Dietary Soy Isoflavones on Estrogen Action in Premenopausal Women.

We are currently investigating the effects of dietary soy isoflavones on estrogen action in premenopausal women. For this project, 19 healthy young women are living at home, consuming 3 different diets for about 3 menstrual cycles each. The 3 diets consist of the subjects' habitual low-fiber diets supplemented with soy powder providing total isoflavones at approximately 0, 1 and 2 mg/kg body weight. The diet periods are separated by 3 week washout periods. A number of estrogen endpoints will be measured, reflecting estrogen availability, metabolism, excretion and action. These include circulating concentrations of estradiol, estrone, progesterone, luteinizing hormone, follicle stimulating hormone and sex hormone binding globulin; urinary estrogen metabolites; fecal estrogens; menstrual cycle parameters; and endometrial histology. This project differs from previously reported studies in that: 1) the subjects are free-living and are consuming soy supplemented ad lib diets; 2) the diet periods are considerably longer; 3) isoflavone levels are controlled; 4) isoflavones are provided relative to individual body weight; and 5) 2 doses of isoflavones are being used in addition to the zero isoflavone control. This study will be completed by the end of 1997; at this point, approximately half of the project has been completed. Preliminary results of the hormone, menstrual cycle and endometrial histology data from the first diet period will be discussed.
